MTDH also known as Metadherin, AGE1(Astrocyte elevated gene-1), LYRIC) is a 582 amino acid single-pass membrane protein that plays a critical role in cancer progression. MTDH is primarily localized to the endoplasmic reticulum membrane, but can translocate from the cytoplasm to the nucleus in response to tumor necrosis factor-alpha (TNFα), highlighting involvement in cellular signaling pathways. MTDH interacts with various proteins, including transcription factor NFκB, which MTDH activates to promote anchorage-independent growth of immortalized astrocytes and melanocytes, essential processes in tumor development. |
