NEDD8 also known as Neddylin is a ubiquitin (Ub)-like molecule that shares 80% homology with ubiquitin, a protein known to modify and target various proteins for proteolytic degradation.NEDD8 plays a crucial role in regulating protein degradation pathways. NEDD8 is primarily localized in the nucleus and is highly expressed in adult heart and skeletal muscle, which is significant for maintaining cellular homeostasis and muscle function. NEDD8 is activated by the E1 ubiquitin-activating enzyme UBA2, forming isopeptide linkages that facilitate attachment to target proteins. NEDD8 is covalently linked to an E3 ubiquitin ligase through the E2 conjugating enzyme Ubc12, preferentially associating with the CUL-2 protein in the E3 ligase complex CBCVHL. This interaction is vital for the regulation of various cellular processes, including cell cycle progression and signal transduction. |
