XPD also known as TFIIH, ERCC2 is involved in transcription-coupled nucleotide excision repair and is an integral member of the basal transcription factor BTF2/TFIIH complex. XPD plays a crucial role in transcription and DNA repair mechanisms, functioning as a part of the TFIIH complex, which is essential for unwinding DNA during transcription initiation and nucleotide excision repair. XPD′s ATP-dependent helicase activity is vital for separating DNA strands, allowing RNA polymerase II to access the template strand for transcription. Mutations in the XPD gene have been linked to various disorders, including xeroderma pigmentosum and Cockayne syndrome, highlighting XPD′s significance in maintaining genomic stability and cellular integrity. |
