MYPT1 also known as PPP1R12A is one of the subunits of myosin phosphatase. MYPT1 plays a crucial role in regulating the interaction between actin and myosin, which is essential for muscle contraction and cellular movement. MYPT1 is primarily localized on stress fibers, where MYPT1 is strategically positioned near the cell membrane and at cell-cell contacts, allowing MYPT1 to effectively modulate myosin phosphatase activity in response to various signaling pathways. MYPT1 is regulated by phosphorylation, particularly at Tyrosine 654 by myotonic dystrophy protein kinase, which influences myosin light chain phosphorylation and smooth muscle contraction. MYPT1′s phosphorylation state is critical, as MYPT1 determines the sensitivity of smooth muscle to calcium, thereby impacting vascular tone and blood pressure regulation.
