nNOS also known as NOS1 belongs to the family of nitric oxide synthases, which synthesize nitric oxide from L-arginine. nNOS is predominantly localized in neurons, where NOS1 interacts with key proteins such as PSD-95, which anchors nNOS to the postsynaptic density, and calmodulin, which regulates enzymatic activity. These interactions are crucial for the precise spatial and temporal production of NO, facilitating NO′s role in modulating synaptic strength and plasticity. Understanding the protein-protein interactions of nNOS is vital for elucidating the mechanisms underlying neuronal signaling and the pathophysiology of neurological disorders where NO signaling may be disrupted.
