MDM4 also known as MDMX is a nuclear protein that contains a p53 binding domain at the N-terminus and a RING finger domain at the C-terminus, and shows structural similarity to p53-binding protein MDM2. MDM4 plays a crucial role in regulating the stability of the p53 protein, a key tumor suppressor involved in cell cycle control and apoptosis. By inhibiting MDM2-mediated degradation of p53, MDM4 ensures that p53 levels are maintained, which is vital for the cellular response to stress and DNA damage. The interaction between MDM4 and p53 is particularly important in the context of cancer, as the amplification of the MDM4 is frequently observed in malignant gliomas, contributing to tumor progression by allowing cancer cells to evade apoptosis. Furthermore, MDM4′s ability to bind and stabilize p73, another member of the p53 family, highlights its multifaceted role in regulating cell survival and death pathways. |
