14-3-3 proteins play a crucial role in regulating numerous cellular processes, particularly those associated with cancer biology, such as apoptosis, mitogenic signaling, and cell-cycle checkpoints. 14-3-3 proteins are located primarily in the cytoplasm and nucleus, where 14-3-3 proteins interact with a wide range of target proteins, influencing their activity and stability. 14-3-3 proteins′ ability to sequester ligands in the cytoplasm is particularly important, as this sequestration prevents ligand degradation and modulates interactions with other cellular components. Such dynamic regulation is essential for maintaining cellular homeostasis and ensuring proper cellular responses to various stimuli. Loss of 14-3-3 expression can lead to dysregulation of these processes, contributing to the development of various diseases, including cancer. |
