Tyk2 is a crucial member of the non-receptor Janus tyrosine kinase family, playing a pivotal role in mediating cellular responses to cytokines and interferons, which are essential for the immune system′s functionality. Located primarily in the cytoplasm, Tyk2 is recruited to activated cytokine receptors, where Tyk2 phosphorylates signal transducer and activator of transcription (STAT) proteins. This phosphorylation triggers the dimerization of STATs, allowing STATs to translocate to the nucleus and regulate the transcription of genes involved in immune responses. Proper functioning of Tyk2 is critical for maintaining immune homeostasis, as dysregulation can lead to various inflammatory diseases. |
