Dab1 also known as SCA37 is a homolog of the Drosophila Disabled protein. Dab1 plays a crucial role as an adaptor protein in neural development, particularly during the rapid expansion of the developing nervous system. Located primarily in the cytoplasm, Dab1 undergoes tyrosine phosphorylation, which is essential for its function in signaling pathways that guide the positioning of neurons. This phosphorylation allows Dab1 to interact with other proteins, notably through its PTB-like domain, facilitating its role in the Reelin signaling pathway. Reelin, a secreted protein, is vital for the proper migration and layering of neurons in the brain, and Dab1 acts downstream of Reelin, mediating its effects. Mutations in Dab1 can lead to significant brain abnormalities, mirroring the defects observed in Reelin mutants, underscoring the importance of Dab1 in maintaining neural architecture and function. |
