p57 also known as Kip2 is a member of the p21 Waf1/Cip1/p27 family of cyclin-dependent kinase inhibitors, playing a significant role in regulating cell cycle progression by inhibiting several G1 cyclin/Cdk complexes. This inhibition is vital for maintaining proper cell cycle control, particularly in response to cellular stress or DNA damage, as p57 prevents premature progression into the S phase, thereby allowing time for DNA repair mechanisms to function effectively. Notably, p57 is not regulated by p53, which distinguishes p57 from other family members like p21 Waf1/Cip1, and p57 overexpression can lead to a robust G1 arrest, highlighting p57′s potential as a therapeutic target in cancer where cell cycle dysregulation is prevalent. |
