NumB plays a crucial role in neuronal cell fate decisions, acting as a signaling adapter with two distinct protein-protein interaction domains: a phosphotyrosine-binding domain and a proline-rich SH3-binding region. This structural configuration is essential for NUMB′s function in mediating interactions with other proteins, such as EPS15, LNX1, and NOTCH1, which are vital for the regulation of cell fate during development. In mammals, NumB homologs are implicated in determining cell fates, and studies have shown that conditional mouse mutants lacking NUMB in developing sensory ganglia exhibit reduced axonal arborization in afferent fibers, highlighting NUMB′s importance in neural development. |
