PSD95 aslo known as SAP90, DLG4 is a member of the membrane-associated guanylate kinase (MAGUK) family, plays a crucial role in synaptic signaling and plasticity by organizing and anchoring various proteins at the postsynaptic density of excitatory synapses. This localization is vital for proper synaptic functioning, as PSD-95 facilitates the clustering of receptors and signaling molecules, thereby influencing synaptic strength and communication between neurons. The structural integrity of PSD-95, characterized by PDZ, SH3, and guanylate kinase domains, allows interaction with various proteins, including NMDA and AMPA receptors, which are essential for synaptic transmission and plasticity.
