c-Myb is a protein with three HTH DNA-binding domains that functions as a transcription regulator. c-Myb is predominantly localized in the nucleus, where c-Myb functions as a critical transcription factor regulating the expression of genes involved in hematopoietic cell proliferation, differentiation, and survival. c-Myb may be aberrently expressed or rearranged or undergo translocation in leukemias and lymphomas, and is considered to be an oncogene. This nuclear localization is essential for c-Myb′s role in controlling transcriptional programs that maintain the balance between cell growth and differentiation, making c-Myb a vital component in normal hematopoiesis and implicated in oncogenic processes when dysregulated.
