Smad1 also known as MADH1, JV4-1, MADR1 is an intracellular signal transducer and transcriptional modulator involved in the Bone Morphogenetic Protein (BMP) pathway, part of the Transforming Growth Factor-beta (TGF-β) superfamily. Upon activation by BMP receptors, Smad1 is phosphorylated and forms a complex with Smad4, translocating from the cytoplasm to the nucleus. This nuclear localization is crucial because Smad1 regulates the expression of target genes that control cell proliferation, differentiation, and apoptosis. Smad1 function is essential for embryonic development, tissue homeostasis, and bone formation. Dysregulation of Smad1 signaling has been implicated in various diseases, including cancer and fibrosis. |
