SirT2 also known as Sirtuin 2 is a member of the silent information regulator (SIR2) family, which is conserved across species and plays critical roles in cellular processes such as transcriptional regulation, DNA repair, and aging. SirT2 characterized by its NAD-dependent deacetylase activity, modulates various cellular functions, including cell cycle regulation and response to DNA damage, making it crucial for maintaining cellular health and genomic stability. SirT2′s catalytic core includes a 323 amino acid NAD-binding domain and a large groove essential for its enzymatic function, facilitating mono-ADP-ribosylation and influencing protein interactions within the cell. Alongside its central role in deacetylating histones and regulating transcription at specific genetic loci, SirT2 is part of a broader network of seven sirtuins (SirT1-7) divided into four classes based on structure and function. |
