SHIP-1 also known as INPP5D is a member of the inositol polyphosphate-5-phosphatase (INPP5) family. SHIP-1 is an essential SH2-containing inositol phosphatase that plays a critical role in the negative regulation of the PI3K/Akt signaling pathway by dephosphorylating PIP3 to PIP2, thereby modulating key cellular processes such as immune cell activation, proliferation, and survival. This regulation is vital for maintaining immune homeostasis and preventing excessive immune responses that can lead to autoimmune disorders and cancer. SHIP-1 interacts with several important proteins, including the CSF-1 receptor (c-Fms), adaptor proteins like Grb2 and Dok-1, and the p85 subunit of PI3K, facilitating the integration and attenuation of various signaling cascades. |
