ATP-citrate synthase also known as ATP-citrate lyase, citrate cleavage enzyme is a cytoplasmic homotetramer that belongs to the succinate/malate CoA ligase family, with its gene located on chromosome 17q12-q21. This enzyme plays a crucial role in cellular metabolism by catalyzing the conversion of citrate and CoA into acetyl-CoA and oxaloacetate. Acetyl-CoA is a vital substrate for fatty acid and cholesterol biosynthesis, making ATP-citrate synthase essential for energy production and lipid metabolism. Additionally, ATP-citrate synthase is significant in the biosynthesis of acetylcholine in nervous tissue, highlighting its importance in neurotransmission and overall brain function.
