CD22 also designated B-lymphocyte cell adhesion molecule (BLCAM), Siglec-2, Leu-14 is a type I integral membrane glycoprotein that plays a crucial role in regulating B cell signaling and maintaining immune homeostasis. CD22 is predominantly expressed on B cells, where CD22 functions as a negative regulator of B cell receptor signaling, thereby preventing excessive activation and promoting self-tolerance. CD22 is located on the cell membrane and is also found in a cytoplasmic form during early stages of B cell development, making CD22 a valuable marker for acute lymphocytic leukemia. Alternative splicing of the CD22 gene results in two isoforms, CD22α and CD22β, with CD22β being more prevalent. CD22 can also exist as a heterodimer composed of CD22β and the shorter CD22α isoform, highlighting the complexity of CD22 regulation and function in the immune response. |
