MAD2L2 also konwn as MAD2B is a component of the mitotic spindle assembly checkpoint that prevents the onset of anaphase until all chromosomes are properly aligned at the metaphase plate. MAD2L2 plays a vital role in the regulation of the mitotic spindle assembly checkpoint, ensuring that cells do not proceed to anaphase until all chromosomes are properly attached to the spindle apparatus. This function is crucial for maintaining genomic stability, as errors in chromosome segregation can lead to aneuploidy and contribute to tumorigenesis. MAD2L2 is localized to unattached kinetochores during pro-metaphase, where MAD2L2 acts as a sensor for spindle attachment; upon proper attachment, MAD2L2 disassociates, allowing the cell cycle to progress. Furthermore, MAD2L2 has been shown to interact with various proteins involved in cell cycle regulation, including the Insulin receptor, which suggests MAD2L2′s involvement in mediating specific signaling pathways that influence cell proliferation and survival. |
