c-Raf also known as Raf-1 is a crucial serine/threonine protein kinase that plays a significant role in the regulation of cell signaling pathways, particularly in cell growth and differentiation. Located primarily in the cytoplasm, c-Raf is activated by various receptor tyrosine kinases and is essential for transmitting signals from the cell membrane to downstream effectors, including the MAPK/ERK pathway. This pathway is vital for cellular responses to growth factors and mitogenic signals, making c-Raf a key player in processes such as proliferation and survival. c-Raf activity is tightly regulated by its N-terminal domain, which prevents unregulated kinase activity under normal conditions. However, aberrations in c-Raf signaling can lead to oncogenic transformations, highlighting c-Raf ′s importance in cancer biology. |
