CHIP also known as STUB1 is a protein containing tetratricopeptide repeat and a U-box that functions as a ubiquitin ligase/cochaperone. CHIP plays a crucial role in protein quality control by mediating the ubiquitination and subsequent degradation of misfolded or damaged proteins. This function is vital for maintaining cellular homeostasis and preventing the accumulation of potentially toxic proteins that can lead to disease. CHIP interacts with molecular chaperones Hsp70 and Hsp90, facilitating the degradation of client proteins such as ErbB2 and CFTR, which is significant in regulating signaling pathways and stress responses. Additionally, CHIP can modulate signaling pathways by interacting with Smad1/Smad4 and inhibiting BMP signaling through the ubiquitin-mediated degradation of Smad proteins. |
