TRIB3 also known as TRB-3, NIPK is a putative protein kinase that is induced by the transcription factor NF-kappaB. TRIB3 plays a critical role in disrupting insulin signaling by directly binding to Akt kinases, thereby inhibiting their activation. This function is particularly important as TRIB3 contributes to insulin resistance, a key factor in the development of type II diabetes. TRIB3 is highly expressed in the liver, pancreas, peripheral blood leukocytes, and bone marrow, with increased levels observed in the livers of diabetic mice compared to their wildtype counterparts. TRIB3 interacts with ATF4, inhibiting its transcriptional activation activity and regulating the activation of mitogen-activated protein kinases (MAPKs). The regulation of these pathways underscores TRIB3′s significance in metabolic processes and potential as a therapeutic target for diabetes-related conditions. |
