Thrombomodulin(TM) also known TRBM, CD141, THBD is a type I membrane receptor that is specific to endothelial cells. Thrombomodulin plays a crucial role in coagulation regulation. Thrombomodulin features a cysteine-rich extracellular domain with six EGF-like regions, which are essential for Thrombomodulin′s interaction with thrombin. This interaction activates protein C, leading to activated protein C (APC) generation, an important anticoagulant enzyme that, with protein S, inhibits coagulation by inactivating factors Va and VIIIa. TM′s significance in maintaining hemostatic balance is underscored by Thrombomodulin gene deletion resulting in embryonic lethality in mice, highlighting Thrombomodulin′s essential role in vascular biology and coagulation homeostasis. |
