TIRAP also known as BACTS1 plays a crucial role in the innate immune response by acting as a signaling adapter that links Toll-like receptors (TLRs) to downstream signaling pathways, including the activation of NFκB and mitogen-activated protein kinases. TIRAP is primarily located in the cytoplasm, where TIRAP forms homodimers and heterodimers with MyD88, facilitating the recruitment of interleukin-1 receptor-associated kinases (IRAKs) to TLRs. Proper functioning of TIRAP is vital for effective immune responses, as TIRAP ensures rapid activation of inflammatory pathways in response to microbial infections. Dysregulation of TIRAP signaling can lead to impaired immune responses or excessive inflammation, highlighting TIRAP′s importance in maintaining immune homeostasis.
