Mnk1 also known as MKNK1 is a Ser/Thr protein kinase that interacts with, and is activated by ERK1 and p38 mitogen-activated protein kinases, and thus may plays a crucial role in cellular signaling pathways, particularly in regulating protein synthesis in response to stress signals. Mnk1 is primarily located in the cytoplasm, where Mnk1 interacts with eukaryotic translation initiation factor 4E (eIF4E), a key player in translation initiation. This interaction enables eIF4E phosphorylation, which enhances affinity for the mRNA cap structure, promoting translation of specific mRNAs critical for cell survival and proliferation under stress conditions. Mnk1′s ability to modulate protein synthesis in response to environmental cues underscores Mnk1′s importance in various physiological processes, including cell growth, differentiation, and stress response, making Mnk1 a significant target for research in cancer and other diseases. |
