RhoGDI also known as RhoGDIα, ARHGDIA is a protein that plays a significant role in regulating Rho GTPases, which are essential for various cellular processes, including cell proliferation and cytoskeletal dynamics. RhoGDI is primarily localized in the cytoplasm, where RhoGDI functions to inhibit the dissociation of guanosine diphosphate (GDP) from Rho proteins, thereby preventing the activation of these GTPases by guanosine triphosphate (GTP). Regulation of RhoGDI is vital, as RhoGDI activity can be modulated by phosphorylation at Ser 101 by p21-activated kinase (αPAK), which can lead to a decrease in inhibitory function and potentially enhance the signaling pathways mediated by Rho proteins.
