S100A4 also known as MTS1 is a member of the S100 family of proteins containing 2 EF-hand calcium-binding motifs. S100A4 plays a crucial role in tumor progression and metastasis, as well as significantly stimulating angiogenesis. S100A4 is primarily located in the cytoplasm and nucleus of various cell types, where S100A4 interacts with multiple signaling pathways that regulate cell growth and survival. S100A4′s presence in these cellular compartments is vital, as S100A4 modulates key processes such as apoptosis and cell migration, which are essential for cancer development and progression. Notably, S100A4 has been shown to cooperate with the tumor suppressor protein p53, binding to its C-terminal regulatory domain and inhibiting its DNA-binding activity, thereby enhancing p53-dependent apoptosis.
