p14 ARF is encoded by the CDKN2A gene, which also produces p16 protein, and interacts with MDM2, a negative regulator of the p53 tumor suppressor pathway. By binding to MDM2, p14 ARF stabilizes p53 levels, leading to increased expression of p21 and subsequent cell cycle arrest in both G1 and G2/M phases. This regulatory mechanism is essential for preventing tumorigenesis, as damaged cells do not continue to divide. p14 ARF plays a crucial role in regulating the cell cycle by acting as a tumor suppressor, which is vital for maintaining cellular integrity and preventing uncontrolled cell proliferation. |
