DLD(Dihydrolipoamide dehydrogenase) also known as PHE3 is a member of the class-I pyridine nucleotide-disulfide oxidoreductase family. DLD plays a crucial role in cellular energy metabolism as a flavin-dependent oxidoreductase, functioning as a key component of several important enzyme complexes, including the pyruvate dehydrogenase and α-ketoglutarate dehydrogenase complexes. DLD is primarily located in the mitochondrial matrix, where DLD exists in various oligomeric forms such as monomers, homodimers, or tetramers, which are essential for enzymatic activity. DLD′s ability to generate NADH and lipoic acid from dihydrolipoic acid and NAD+ is vital for maintaining the redox balance within the mitochondria, thereby supporting ATP production and overall cellular function. |
