IRS-2(Insulin Receptor Substrate 2) plays a crucial role as a signaling intermediate downstream of several receptors, including insulin, insulin-like growth factor 1, interleukin-4, interleukin-9, and interleukin-13.iRS-2 is involved in activating mitogen-activated protein kinase (MAPK) and protein kinase B (PKB) pathways, which are essential for cellular responses to growth factors. In IRS-2-deficient mice, a significant reduction in total phosphoinositide 3-kinase activity and subsequent loss of protein kinase C (PKC) zeta activation can lead to type 2 diabetes, highlighting IRS-2′s importance in metabolic regulation. IRS-2 translocation to nuclei of mouse embryo fibroblasts expressing insulin-like growth factor 1 receptor underscores its multifaceted role in cellular signaling and gene expression.
