FAT10 also known as Ubiquitin D, Diubiquitin is a 165 amino acid protein encoded within the major histocompatibility complex (MHC) and features two domains that exhibit significant homology with ubiquitin. Each domain contains two cysteine residues and a free C-terminal diglycine motif, which is essential for the formation of FAT10 conjugates. FAT10 is notably inducible by interferon-γ and tumor necrosis factor alpha (TNFα), highlighting its role in immune responses. FAT10 is crucial for various cellular processes, including antigen presentation, cytokine response, apoptosis, and mitosis, and interacts with MAD2, a key component of the spindle checkpoint, thereby influencing cell cycle regulation. |
