PAX8 is a member of the paired box (PAX) family of transcription factors. PAX8 plays a significant role in embryogenesis and differentiation of thyroid follicular cells, vital for proper thyroid hormone biosynthesis. PAX8 is predominantly expressed in developing and adult thyroid, developing secretory system and, to a lesser extent, adult kidney. PAX8 interacts with other transcription factors, such as TTF-1 and TTF-2, to regulate expression of key genes involved in thyroid function, including sodium iodide symporter (NIS), thyroid peroxidase (TPO), thyroglobulin (TG), and thyrotropin receptor (TSHR). Mutations in PAX8 can lead to thyroid dysgenesis, highlighting its importance in thyroid development and function.
