NFS1 also known as nitrogen fixation 1 is a crucial enzyme that plays a significant role in the biosynthesis of iron-sulfur (Fe-S) clusters, which are vital cofactors for numerous biological processes. NFS1 is primarily located in the cytoplasm and mitochondria, with localization being influenced by cytosolic pH levels. This dual localization allows NFS1 to participate in various metabolic pathways, including the synthesis of Fe-S clusters that are essential for electron transfer in oxidative phosphorylation and enzymatic reactions in key metabolic enzymes such as aconitase and dehydratases. NFS1′s ability to supply inorganic sulfur for Fe-S cluster assembly underscores its importance in maintaining cellular function and integrity, particularly in energy metabolism and DNA repair mechanisms.
