NK-1R also known as TACR1 is a member of the tachykinin receptor family, which includes NK-2 and NK-3 receptors, and plays a significant role in various physiological processes. NK-1R is primarily located in the central nervous system, particularly in the dorsal horn of the spinal cord, where NK-1R is expressed in a high proportion of spinothalamic and spinobranchial neurons. NK-1R localization is crucial as NK-1R is involved in nociceptive transmission, influencing pain perception and the modulation of chronic neuropathic and inflammatory pain. Furthermore, NK-1R′s interaction with tachykinin peptides, such as substance P, substance K, and neuromedin K, highlights NK-1R′s importance in mediating responses related to anxiety and depression.
