NKCC2 also known as SLC12A1 is a kidney-specific sodium-potassium-chloride cotransporter that is expressed on the luminal membrane of renal epithelial cells of the thick ascending limb of Henle's loop and the macula densa. NKCC2 plays a crucial role in the kidney, specifically in the thick ascending limb of the loop of Henle, where NKCC2 is responsible for active reabsorption of sodium and chloride ions. This function is vital for maintaining electrolyte balance and blood pressure regulation in the body. The gene encoding human NKCC2 is located on chromosome 15q15-q21, and mutations in this gene can lead to Bartter′s syndrome, a genetic disorder characterized by hypokalemic alkalosis and metabolic imbalances. NKCC2 is sensitive to diuretics such as furosemide and bumetanide, which are commonly used in clinical settings to manage conditions like hypertension and edema.
