GR (glucocorticoid receptor) also known as NR3C1 is a critical transcription factor that plays a vital role in mediating the physiological effects of glucocorticoids, which are steroid hormones involved in numerous biological processes such as metabolism, immune response, and stress regulation. GR is predominantly located in the cytoplasm, where GR exists in an inactive form bound to heat shock proteins, particularly HSP90. Upon binding to glucocorticoids, GR translocates to the nucleus, where GR regulates gene expression by either inducing or repressing target genes. This translocation allows GR to interact with specific DNA sequences and recruit co-regulators that modulate transcription. GR′s ability to influence gene expression is essential for maintaining homeostasis and responding to environmental changes, making GR a significant target for therapeutic interventions in conditions such as inflammation, autoimmune diseases, and cancer. |
